要旨
Most metabolites involved in the primary energy metabolism are anions; however, their concentrations in cells and tissues are often too low to be measured accurately by conventional metabolomics platforms. Here, we developed a capillary electrophoresis-triple quadrupole mass spectrometry (CE-QqQMS)-based metabolomics technique, which dramatically improved detection sensitivity of 63 anions up to 291-folds compared to a conventional analysis. The method was then applied to characterize metabolomic profiles of paired non-tumor and tumor tissues of 27 gastric cancer patients. The results not only distinguished their profiles but also delineated differentiation-status-dependent tumor profiles: Differentiated tumor (DT) tissues showed considerably high glycolytic intermediate, nucleotide, and glutathione levels, whereas undifferentiated tumor (UT) tissues rather exhibited relatively low energy and redox statuses, which were also observed in DT and UT cell lines. Taken together, DT exhibited more typical cancer metabolic profiles than UT represented by active Warburg- and amino acid-dependent metabolism, whereas low glutathione ratios in UT imply their vulnerability to oxidative stress.

要旨
Backgrounds: Metabolomic characteristics of esophageal cancer have been largely unknown. We thus conducted metabolome analysis of paired non-tumor (NTs) and tumor (Ts) esophageal tissues by using capillary electrophoresis time-of-flight mass spectrometry (CE-TOFMS).Methods: Tumor and surrounding non-tumor tissues were surgically excised pair-wise from 10 esophageal cancer patients. Following tissue homogenization and metabolite extraction, we measured 235 metabolites by CE-TOFMS.Results: Principal component analysis showed clear metabolomic differences between NTs and Ts. Lactate and citrate levels in Ts were significantly higher and lower, respectively, than those in NTs, which reflects hypoxic microenvironment and corroborates the Warburg effect in Ts. Concentrations of most amino acids except Gln were higher in Ts than in NTs presumably due to accelerated glutaminolysis and autophagic protein degradation in Ts.Conclusion: The hallmarks of cancer metabolism were reassured by comparing metabolomic profiles of esophageal NTs and Ts. Several metabolites were detected only in tumor tissues, which may lead to elucidate tumor-specific metabolic pathways.
